The Ocular Research group of Spark Therapeutics is seeking a highly motivated Research Associate to perform viral vector potency and mechanistic of action studies in support of ocular programs. Key to this position is demonstrated expertise in primary cell culture, iPSC derived organoid cultures and/or 3D culture systems. The candidate will be well versed in cell culture to facilitate rapid and accurate assessments of viral vector potency.

 

Primary responsibilities:

 

  • Develop cell culture systems for potency and biological responses to novel vectors
  • Implement new cell-based assays or optimize current assays based on in-depth knowledge of laboratory techniques.
  • Quantify transgene levels and correlate with protein product using a variety of routine assays (eg. molecular cloning, cell culture, transient transfections, AAV transduction, DNA/RNA extraction, ELISA, qPCR, western blot, immunohistochemistry, etc.)
  • Participate in sample management activities such as receiving, processing, storage and shipping preclinical samples.
  • Accurately document assay results and analyze data using an electronic lab notebook, technical documents, and other reporting formats. 
  • Contribute to a collaborative, creative and rigorous culture of scientific discovery

 

Responsibilities:

70%  Plan, prepare and perform experiments on the bench to support the Ocular Platform research projects. Assist technical staff in the performance of laboratory experiments as needed.

20%  Analyze results, prepare technical reports, and accurately document work in an electronic lab notebook. Prepare Powerpoint presentations of results as needed. Communicate results and research progress during team meetings as needed.

10%  Participate in group meetings, scientific discussions, and provide support to more junior staff, as needed.

 

Requirements:  

  • BA/BA in Biological sciences required, Master’s in a relevant field preferred.
  • 3-5 years of laboratory experience.
  • In-depth knowledge of mammalian cell culture techniques including primary cell lines, tissue explant culture, organoids, cell characterization/differentiation, transient transfection, and viral transduction.
  • Proven hands-on experience with cell-based and molecular biology assays with a variety of readout technologies, such as but not limited to cell culture, flow cytometry, DNA/RNA/protein extraction, qPCR, ELISA, luciferase and/or enzymatic activity assays and Western analysis. Immunohistochemistry/FISH and Jess/Wes experience is a plus.
  • Experience with AAV vectors is a plus.
  • Experience with tissue dissection is a plus.
